We tested the effects of doxycycline on the expression of genes related to the immune system and reproduction. Methods

We performed RNA isolation and real-time PCR (RT-PCR) of the selected genes, and assessed the effects of doxycycline on their expression levels. RNA was extracted from the cerebrum and testis using the QIAamp Viral mRNA Extraction Kit (Qiagen, Valencia, CA, USA). Reverse transcription was carried out for 40 min at 70°C and the mRNA was isolated using the MagPran mRNA Kit (Thermo Fisher Scientific, Waltham, MA, USA). The sequences of the cDNAs were used to construct the cDNAs. All primer and probe sequences used are listed in Supplementary Table.

All the genes whose expression was assessed by real-time RT-PCR were normalized to the expression levels of the reference genes Acta2, Actin, and GAPDH. The primers are listed in Supplementary Table.
